Sinopsis

Conférence - Emmanuelle Passegué : Hematopoietic Stem Cell Aging and Adaptation to Stress
18/11/2025 Duración: 01h05minHugues de ThéCollège de FranceOncologie cellulaire et moléculaireAnnée 2025-2026Hematopoietic Stem Cell Aging and Adaptation to StressConférence - Emmanuelle Passegué : Adaptive and Maladaptive Myeloid Cell ProductionEmmanuelle PasseguéDirectrice de la Columbia Stem Cell Initiative et professeure émérite de génétique et de développement à l'université ColumbiaRésuméAlthough young adult HSCs employ finely tuned mechanisms to balance pro-survival and stress responses, these maintenance strategies provoke vulnerabilities that manifest with continued age, leading to a decline in function. We will examine HSC biology through the lens of antagonistic pleiotropy, whereby the same mechanisms that are important for reproductive fitness form the basis for functional decline during aging. We will particularly highlight the central role of cell cycle regulation, niche cell reliance, inflammatory responses, cellular memory, and distinct metabolic regulation in driving HSC aging features, including expansion of the HSC poo

We will consider how acute insults, such as infections and myeloablation, cause molecular, cellular, and metabolic changes in hematopoietic stem and progenitor cells at multiple levels of the hematopoietic hierarchy to drive accelerated production of mature myeloid cells needed to resolve the initiating insult. Moreover, we will discuss how dysregulation or subversion

Interactions among stromal cells and hematopoietic stem and progenitor cells modulate many of the critical functions of the hematopoietic system, including production of specific blood lineages and responses to infection and inflammation. Our understanding of these interactions has grown considerably with application of new technologies that have uncovered how cell populations associate in situ and exchange molecular signals. We

Self-renewing HSCs reside at the apex of this hierarchy and produce all blood lineages, including the erythroid lineage that forms red blood cells, the megakaryocytic lineage that gives rise to platelets, and the myeloid and lymphoid lineages that generate cells of the innate and adaptive immune systems. Most HSCs exist in a quiescent state, dividing occasionally to self-renew and produce various lineage-b

Conférence - Tony Hunter : New Insights into Intercellular Crosstalk between Different Cell Populations in Pancreatic Tumors That Can Be Translated into the Clinic
27/05/2024 Duración: 59minHugues de ThéCollège de FranceOncologie cellulaire et moléculaireAnnée 2023-2024Exploring the World of Protein Post-translational ModificationsConférence - Tony Hunter : New Insights into Intercellular Crosstalk between Different Cell Populations in Pancreatic Tumors That Can Be Translated into the ClinicTony HunterSalk Institute, La Jolla, California, USARésuméPancreatic adenocarcinoma (PDA) is one of the most lethal cancers, primarily due to late diagnosis. PDA is a highly stromal (desmoplastic) and poorly vascularized tumor with a dense extracellular matrix; only ~10% of the cells in the tumor are tumor cells, with the rest of the tumor microenvironment (TME) being comprised of cancer-associated fibroblasts (CAFs, and immune cells, including tumor-associated macrophages (TAMs). To investigate the role of cellular crosstalk between CAFs and tumor cells TME, we set out to identify paracrine factors secreted by activated CAFs in the tumor tissue that act on the tumor cells and can be targeted for therapeutic
